چکیده

The expression levels of the peripheral blood mononuclear cells surface Toll-like receptors (TLR2, TLR4), the quantity of B lymphocyte subgroups, and the number of apoptosis lymphocyte in peripheral blood were detected to investigate their implication in the occurrence and development of ankylosing spondylitis and propose a new system for monitoring patients and therapeutic strategies. We analyzed the peripheral blood specimens stemming from 67 Ankylosing spondylitis (AS) patients and 30 healthy individuals through flow cytometry. Two reagents, FITC TLR2 and PE-TLR4, were used for the detection of expression levels of TLR2 and TLR4 while APC-CD19 and PE-CD5 were used in the enumeration of different B lymphocytes subgroups. For the detection of lymphocytes apoptosis, we employed FITC and streaming Annexin-v/PI method. The expression levels of peripheral blood mononuclear cells surface TLR2 and TLR4 appeared unusually high in the patients with AS and was not associated with disease activity. Similarly, there were abnormal changes in the amount of B lymphocytes subset in the patients with AS. The amount of apoptosis lymphocytes in peripheral blood were reduced in AS patients which was correlated with the abnormal increase of CD19+ and CD19+CD5+ B lymphocytes. Apoptosis Lymphocytes as well as the amount of CD19+ and CD19+CD5+ B lymphocytes were all associated with AS disease activity. The present study provided new insights into the physiopathology and the pathogenesis of AS disease and suggested new strategies for the monitoring and treatment of AS patients.

Simvastatin and Recombinant Antagonist of Receptors of Interleukin-1 Modulate Toll-like Receptors in Experimental Acute Ileitis in Rat

Background: The pathogenesis of inflammatory bowel disease is complex and multifactorial. Studies have led to the current concept that Toll-like receptors represent key mediators of innate host defense in the intestine, and they are involved in maintaining mucosal as well as commensal homeostasis. We studied the possibility of simvastatin and antagonist of receptors of interleukin-1 for pharmac...

P-17: Expression of Cell Surface Toll-Like Receptors in the Human Male Reproductive Tract

Background: Male infertility refers to the inability of a male to achieve a pregnancy in a fertile female. The root of many causes of infertility is miscommunication between immune and reproductive system. Male reproductive system is very sensitive and vulnerable, infections can hinder maturation and movement of spermatozoa lead to impaired fertility. All species need an immediate reply to the m...

TLR2 and TLR4 Signaling Pathways and Gastric Cancer: Insights from Transcriptomics and Sample Validation

Background: Pattern recognition receptors, especially toll-like receptors (TLRs), as the first line of defense for pathogen detection, were found to be associated with H. pylori infection and gastric cancer (GC). However, the expression levels of TLRs, i.e. TLR2 and TLR4, as the main receptors sensed by H. pylori, still remain largely ambiguous. We aimed to investigate the patterns of key tra...

Inhibitory Killer Cell Immunoglobulin-Like Receptor KIR3DL1 in Combination with HLA-B Bw4iso Protect against Ankylosing Spondylitis

Background: The HLA class I molecules serve as ligands for both T cell receptors and killer cell immunoglobulin-like receptors (KIRs). Objective: We investigated the HLAC and HLA-Bw4 alleles as well as KIRs expression on CD56 positive lymphocytes to evaluate whether these genes and molecules could influence Ankylosing spondylitis (AS) susceptibility, alone or in combination. Methods: We typed 4...

Activation of Toll-Like Receptors 2 by High-Mobility Group Box 1 in Monocytes from Patients with Ischemic Stroke

Background: Stroke is a leading cause of death all around the world, and ischemic stroke is considered to be the most common stroke type. Toll-like receptors (TLRs) are important molecules for detection of both pathogen invasion and tissue damage. In this regard, the purpose of this study was to assess the expression level of TLR2 on monocytes in patients with ischemic stroke and to evaluate th...
